Abstract/Description: | Extraction of good quality DNA is the most essential requirement for molecular analysis of any species. Five protocols for DNA extraction were compared to assess their effects on amount of DNA recovered, its purity and efficacy in PCR amplification using RAPD, ISSR and SSR markers in two species of Dendrocalamus as per and DhamiltoniiLeaftissuesfromin itroculturestissuecultureraisedplantsingreenhouseandyoungandmatureleavesfromfieldgrownplantsofthetwospeciesweresubjectedtofive DNA extractionmethodsusingtheprotocolsSignificant differences in DNA quantity and quality between samples and protocols were observed. Leaves of tissue culture raised plantsgrowinginthe green house gave best DNA yield (5681, 5015 ng/μlfollowedbyin) vitroshootcultures(4645, 4379 ng/μl), and young (2720, 1801 ng/μl) and mature leaves (1800, 1368 ng/μl) from field grown plants of D. hamiltoniiandD. asperrespectivelyTheconcentrationofDNAobtainedwasmoreforD. hamiltoniisamplesthanthecorrespondingD. asper tissues. |
