Abstract/Description: | Pseudomonas glumae [Burkholderia glumae] EM85 (Afu+Sid+Flu+HCN+), a potent biocontrol agent for phytopathogenic Rhizoctonia solani of cotton, was monitored using Tn5::lacZ molecular marker. The introduction of lacZ into the chromosome of EM 85 resulted in deficient and overproducing mutants of different antifungal traits. Fluorescent pigments and antifungal antibiotics were responsible for biocontrol of R. solani. Colonies of wild type, isogenic, deficient and overproducing mutants were selected using Lac+ phenotype, antibiotic resistance markers and fluorescence. A comparison of wild type and mutants in terms of their abilities to colonize the root zones of cotton revealed that in general, deficient mutants (Afu-, Sid-, Flu-, HCN- etc.) built up higher population than did the wild type, isogenic and overproducing mutants. This indicated that mutational inactivation or overproduction of antifungal traits did not affect the establishment, population dynamics and ecological fitness of P. glumae EM85. |
