Abstract/Description: | Puntius sophore is an important food fish and an ornamental fish having international demand. The present study examined partial cytochrome b sequence of mitochondrial DNA to assess the genetic differentiation in three populations (Loktak Lake, Langjing Achouba and Lokchao River) of P. sophore in Manipur, North East India. The partial cytochrome b gene (307 bp) revealed one variable site and one parsimony informative site. A total of two distinct mtDNA haplotypes were identified in three populations of P. sophore. The nucleotide diversity for the all the three populations was found to be 0.00133 and haplotype diversity (Hd) was 0.409. The nucleotide diversity for Loktak Lake, Langjing Achouba and Lokchao River populations were 0.000, 0.00163 and 0.00217 respectively. The within population variation accounted for 93.33% of total variation and while 6.67% was attributed to among population variation. The total fixation index (FST) value, 0.06667 was found to be insignificant at P>0.05. The cytochrome b gene sequences revealed low level of genetic differentiation within and between populations of P.sophore and confirmed the suitability of partial cytochrome b gene (307 bp) sequence in determining the genetic diversity in P.sophore. |
