Abstract/Description: | In India, several guava (Psidium guajava) cultivars have emerged as a result of seedling selection and seedling of these cultivars are being commercially exploited through seed propagation and has indirectly given rise to several types which are not true to the commercial type and vary in several character from the parent population. Based on the morphological characters it is not always possible to discriminate between closely related guava genotypes although several morphological markers like fruit colour, leaf shape and size may be useful but they lose their usefulness for discriminating between very closely related guava genotypes. Keeping above in view the efficacy of molecular (DAMD) vis-à-vis morphological parameters was tested to discriminate between individuals in half sib population of Psidium guajava consisting of 6 half-sib progeny (CISH-G-1, G-2, G-3, G-4, G-5 and G-6), Allahabad Safeda and 2 Psidium species. In order to detect adequate levels of polymorphism between these genotypes, a total of 6 minisatellite primers were screened and 4 primers that gave best results in terms of discrete and consistent band patterns were selected. Analysis of the profile obtained after amplification of the guava genome with four primers namely HVR, HBV, M13 and 33.6b depicted that the half-sib seedlings could be discriminated from Allahabad Safeda, P. acutangula and P. guinense. The genetic distance among the half-sib progeny was less but they showed considerable distance from Allahabad Safeda and the two Psidium species studied. |
