Description Light-microscope examination was performed on sections of seeds of the desi cultivar H208 and the kabuli cultivar 501 20-50 days after flowering. In kabuli seeds, the epidermis develops into a uniseriate palisade layer without thickening of the cell wall. In desi seeds, it develops into a multiseriate palisade layer which later develops into thick-walled sclereids which are heavily stainable with toluidine blue, indicating the presence of phenolic compounds contributing to testa colour. The walls of the subepidermal cells do not thicken in kabuli seeds, even though their size is considerably reduced. In desi seeds, these cells become thick walled as the seed matures. It is concluded that permeability and germinability of the seeds may be considerably influenced by the observed differences in the structure of the palisade layers of the testas of desi and kabuli types
