Description Achyranthes aspera is an important medicinal plant. In the present study genetic variability between different population of A. aspera and bioefficacy of extracts of this plant has been studied. Hypothesis: Genetic variation within and between plant population is an important parameter to be determine the level and structure of genetic variation within plant species including evolutionary history. Isozyme analysis is one of the basic approach to determine genetic variation. The traditional methods for treatment of diseases is the practices used by the local people throughout the country. Indian system of medicine have identified 1500 medicinal plant. Keeping in view of A. aspera an important medicinal plant in traditional system, it has been chosen for further study to validate the claim made in traditional system. Methodology: Achyranthes aspera plant have been collected from nineteen district of Haryana. A total of 500 plants were taken for study genetic variability by PAGE following the standard methods. To study the medicinal value of the plant the different plant parts extracts of the plant were prepared from locally collected plants in different solvents. The antibacterial activity was studied by disc diffusion method against gram positive and gram negative bacteria. Anti-inflammatory, anti-hyperglycemic, hypolipidemic and antitumor activity tested at CDRI, Lucknow by following the standard protocols. The statistical analysis for study of genetic variability was followed from Nei (1972), Ferguson (1980), Zar (1974) and Microsoft Excel. Observation and Results: The banding pattern at 5 gene enzyme system (EST, ACPH, CAT, PGM and SOD) have led to 15 genetic polymorphic loci. Gel slices stained of EST-2, EST-3, ACPH-1, ACPH-2, ACPH-3, PGM-1, PGM-2, PGM-4 and SOD-1 consisted of two-banded pattern representing heterozygotes (FS) genotypes. The occurrence of two-banded heterozygotes revealed monomeric enzyme at these locus. CAT-2 consisted of single-banded and two-banded pattern representing homozygotes (SS)
