Seed caffeine content of the diploid species of Coffea L., viz. C. liberica, C.canephora, C. salvatrix, C. eugenioides, C. racemosa and Psilanthus bengalensis, the tetraploid descendants of the interspecific hybrids, Ligenioides (Amphiploid of C. liberica x C.eugenioides) and Racemusta (spontaneous tetraploids derived from diploid Fl of C. racemosa x C. canephora) and further descendants derived by crossing Ligenioides with the Arabicoid Hibrido de Timor and C. canephora with Psilanthus travencorensis and P.wightiana was studied by HPLC technique. From the results it is evident that the genes conditioning caffeine content were acting as co-dominant alleles in the diploid interspecific hybrids. Caffeine data confirms the parthenogenetic descent of the progenies of crosses C. canephora x Psilanthus travencorensis and C. canephora x Psilanthus wightiana. In the tetraploid crosses and backcrosses, the caffeine content is stable suggesting that reduction in caffeine content of C. arabica is possible only by using another tetraploid with low caffeine and appropriate selection in the progenies. Backcross breeding of C. canephora and Mozambicoffea species hybrids to evolve Robusta (C. canephora) with low caffeine is believed to be possible. Tetraploids emerging in these breeding exercises are proposed to be utilized to advantage in breeding C.arabica
