Description |
Conventional methods of enumeration of yeasts and moulds in foods and feeds involve long incubation periods (5 days) at ambient temp. A rapid method was evolved using a more nutritious medium (DYPBS medium) consisting of malt extract or yeast extract, beef extract, dextrose, peptone and NaCl (pH 6.1) and incubation at 30 degree C for 48 h. Chlorotetracycline and chloramphenicol at 100 p.p.m. levels were used for controlling bacterial growth. Comparative studies carried out using 2 storage fungi (Aspergillus sp. and Rhizopus sp.) and 3 strains of yeasts (Saccharomyces cerevisiae, Hansenula anamola and Torulopsis holmii) with 6 different conventional media indicated better recovery using the DYPBS medium. Recovery of added Aspergillus niger spores to different food grains was high with DYPBS medium, and compared to other media gave highest recoveries from rice, wheat, Bengal gram and sorghum. The modified method was also found to help in reviving cultures that had been subjected to heat, chill, freeze or osmotic stress. [From En summ.]
