Description |
Rhodotorula gracilis (ATCC-90950) was found to secrete an extracellular chitosan hydrolysing enzyme constitutively, but was incapable of growth on chitosan as carbon source. The enzyme production appeared to be growth associated in all the three media (Minimal salts + peptone, Yeast extract-peptone-dextrose and potato dextrose) tested. In the potato dextrose broth a peak level of enzyme was found 30 h of culture, after which there was apparent inactivation of enzyme. Such a pattern of enzyme loss was not noticed in the other two media. The organism could grow on glucose, sucrose, xylose, succinate, glycerol or mannitol, each employed as sole carbon source and produced chitosanase with these substrates. Enzyme was produced in ammonium sulphate, ammonium chloride, sodium nitrate, urea as nitrogen source, but not in ammonium nitrate. Several amino acids also supported chitosanase production. |
