Description Black nightshade (Solanum nigrum) belongs to family solanaceae which is commonly known as makoy, deadly nightshade, etc. and is a potential herbal alternative as anti-diabetic agent and various other medicinal properties. The present study describes the extraction of bioactive compounds from leaves of Solanum nigrum , identification and quantification of organic acids present and its beneficial effects on diabetic status in rats. The fresh leaves of S.nigrum were extracted with water and ethanol as solvents using conventional extraction and microwave extraction. A higher anti-inflammatory activity was observed in extract obtained from microwave extraction (64.23 %) than obtained from conventional method (23.75 %). This extract was used for in vitro and in vivo analysis of antidiabetic activity. Identification and quantification of bioactive compounds present was analysed by HPLC. It was observed that in 1 gram of fresh leaves, 1.74 mg of coumaric acid, 1.08 mg of quercitin, 0.82 mg of catechol, 0.52 mg of caffeic acid, 1.06 mg of gallic acid and 0.46 mg of protocatechnic acid. The extract showed a significant inhibition of α-amylase (73.15%), α-glucosidase (64.5%) and glucose diffusion (68.67%). The % inhibition was found to be dose dependent. The beneficial effect of Solanum nigrum on diabetic status was studied on different parameters such as weight loss, blood glucose level, water intake, diet intake, urine volume, urine sugar and lipid profile. The groups treated with high and medium dose of extract showed significant improvement while the low dose treated group showed only marginal improvement.
