Description In the present study an attempt has been made to screen plants/plant parts and fungi
(basidiomyces) aqueous extracts for the ability to degrade AFB1, and to develop a molecular
method through which aflatoxin detoxification can be detected.
Aqueous extracts obtained from plants, plant parts and basidiomycetes fungi were
evaluated for their ability to degrade aflatoxin B1 by thin-layer chromatography and competitive
indirect enzyme-linked immunosorbent assay (ciELISA). Of the various plant extracts, the
extract of Mint (Mentha arvensis) showed the maximum degradation up to 50 %. Followed by
Brahmi (Centella asiatica). Extracts of yucca (Calotropis gigentea) and soybean (Glycine max)
seeds showed shift in mobility of AFB1 on TLC. Among the eight aqueous extracts of
basidiomycetes fungi analyzed, Armillaria sp. and Ganoderma sp. showed complete degradation
as analyzed by TLC and ELISA.
A molecular method for the detection of AFB1 detoxification is under progress. Initially
AFB1 was treated with λDNA in the presence of S9 ezymes. Further AFB1-DNA adduct was
purified and further treated with various restriction enzymes. Restriction pattern was analyzed
for control λDNA and AFB1-DNA adducts λDNA. Initial studies showed that, restriction
enzyme Hae III, Alu I and Bam H1 were non sensitive to AFB1-DNA adduct, hence they showed
normal kind of restriction pattern. Not I was not having any restriction site in λDNA and Sma I,
being a methylation sensitive enzyme, showed varied results. Screening of AFB1-DNA adduct
sensitive restriction enzymes is under progress.
