Description |
This study aims to develop polymer-lipid nanoencapsules of lutein to increase lutein stability and bioavailability. Lutein was extracted and purified using open column chromatography and the purity of lutein was found to be 97 ± 2%. Polymer-lipid lutein nanoencapsules were prepared with varying concentrations of polymer and combinations of surfactants. It is evident that among various concentrations (2.5 mg – 100 mg) of PLGA different types of surfactants and different ratios of PLGA (85:15, 75:25, and 50:50) resulted in varying degrees of particle size distribution. PLGA concentration (2.5mg) and the surfactant. Poly vinyl alcohol and PLGA ratio (50:50) showing better result with respect to particle size and zeta potential of nanoparticles. Hence for the detailed studies, 2.5 mg of PLGA (50:50) and 1 mg of lutein with PVA surfactant were used. Further, results also demonstrate that the PLGA concentration and the size of the particles are directly proportional. Particle size ranged between 93-208 nm which was further confirmed by Scanning electron microscopy, Atomic force microscopy. Bioavailability of lutein in vitro showed that Lutein-PLGA-lipid nanoencapsules was significantly higher (18.91%) than control. PLGA thus may serve as novel carrier for enhancing the lutein bioavailability.
