Description To assess the cytological and cyto-histological causes
associated with male sterility in rice, two cytoplasmic male
sterile lines viz., MS 577A and IR 58025A along with theirĀ· corresponding
maintainer lines (MS 577B and IR 58025B) and a restorer
line (Vajram) were studied in detail during microsporogenesis and
microgametogenesis using light microscopey adopting standard
cyto-histological methods of investigation.
The cyto-histological studies revealed that the behaviour of
anther sac layers of both the male sterile lines were not
different from that of their corresponding B lines and the R
line. The degeneration of anther sac layers in the CMS lines
initiated at the early microspore stage and finally disintegrated
to facilitate pollen dehiscence as in the normal fertile lines
without any hypertrophy.

No abnormal meiotic behaviour was observed in CMS lines t:t..at
are comparable to their maintainer and restorer lines. some
morphological variations in anther length, width and pollen
diameter were observed between the male sterile lines and their
corresponding maintainers and the restorer line.
Some deviations in the mechanism of pollen breakdown was
observed in the tested CMS lines. In IR 58025A, the critical
stage of breakdown was found to be occur at binucleate stage
while in MS 577A, pollen grains are morphologically similar to t~e
corresponding 'B' line but failed to fertilize on selfing.
Based on the above findings, the following conclusions are
derived.
1. The behaviour of the anther sac layers was normal in both
the male sterile lines.
2. Meiosis was also normal in both the CMS lines.
3. Morphological variations are observed between anthers of
male sterile lines and their corresponding maintainer and
restorer lines.
4. In IR 58025A, the critical stage of breakdown was at
binucleate stage whereas in MS 577A, pollen grains were
morphologically similar but failed to fertilize on selfing and
the probable reason for this breakdown of pollen may be due to the
incompatibility between nuclear and plasma genes.
5. It is suggested to initiate future studies at a biochemical
level to specify the breakdown of CMS lines very precfsely.
