A total of six pathogens comprising Escherichia coli (2), Staphylococcus aureus (2) and Proteus sp. (2) were isolated from urine samples of urinary tract infection (UTI) patient. They were examined for antibiotic and serum resistance. E. coli BJ 83, coagulase positive S. aureus and Proteus sp. showed resistance to various antibiotics viz., ampicillin, cefuroxime, streptomycin, etc. as well as resistance to 2% human serum. Amongst them, E. coli BJ 83, which exhibited 62.5% resistance to various antibiotics and serum, was selected for genetic evaluation as well as cotransformation studies. Plasmid curing showed the location of antibiotic resistance markers on R plasmid. The presence of MDR plasmid in E. coli BJ 83 was confirmed by performing RFLP. E. coli BJ 83 plasmid DNA profile showed λmax at 260 nm and Tm of 90ºC. Coexpression of ampicillin and serum resistance by recipient strain of E. coli X-239, after plasmid transformation, confirmed that they are found to be linked markers. |
