155 Fungal isolates, isolated from marine water, belong to five fungal species viz Aspergillus niger, A. fumigatus, Penicillium expansum, Trichoderma virens and Penicillium spp were screened for alpha amylase production. All the isolates belong to A.niger, 12 isolates of Trichoderma virens and A.fumigatus, 10 isolates of Penicillium expansum and 2 isolates of Penicillium spp , were produced alpha amylase. A.niger strain SK01 was selected for alpha amylase production. Vegetables wastes were used as a substrate by solid state fermentation under 0,10,20,40 and 50% moisture content. Enzyme activity was studied at 30°C, 40°C, 50°C, 60°C, 70°C temperature and the pH 5,6,7,8 and 9. Wastes supported growth and enzyme activity at all parameters tested. Maximum activity was recorded at 0% moisture content, 70°C temperature and pH 9. |
