The present study was conducted to assess the effect of insulin and Ovsynch on induction of oestrus in anoestrus buffaloes. Total 30 healthy anoestrus buffaloes were selected and divided into three equal (n=10) groups. Group 1 buffaloes were treated with single injection of biphasic isophane r-DNA insulin hormone (Human Mixtard) subcutaneously @ 0.25 IU/kg b.wt. daily for 3 consecutive days (i.e. -3, -2, and -1 day). Buffaloes from Group 2 were treated with insulin similar to Group 1 for 3 consecutive days followed by a single intramuscular injection of GnRH 10 μg/animal on day 0, injection of PGF2α @ 25 mg/animal on day 7 and second injection of GnRH with same dose on 9th day. Animals of Group 3 (Ovsynch protocol) were treated with single intramuscular injection of GnRH 10 μg/animal on day 0, injection of PGF2α @ 25 mg/animal on day 7 followed by the second injection of GnRH on 9th day. All the treated buffaloes were observed for the oestrus twice daily in morning and evening hours for oestrus behavior and also with per rectal examination once in a day. Buffaloes detected in oestrus were inseminated twice at the 12 hrs. interval with frozen semen after the onset of oestrus. Pregnancy diagnosis was done at 60 days after artificial insemination. Rate of induction of oestrus was higher in insulin + Ovsynch protocol group (G-2, 70.00%) as compared to the insulin alone (G-1, 60.00%) and Ovsynch protocol (G-3, 50.00%). The time required for the onset of estrus in insulin treatment group (G-1) was 14.75 ± 2.78 days (354.00±66.79 hrs.) and in insulin+Ovsynch protocol group (G-2) it was 12.00 ± 0.425 days (288.00±10.14 hrs.). In Ovsynch protocol group (G-3) it was 9.70 ± 0.12 days (232.80±2.93 hrs.) and when calculated from dose of PGF2 it was 56.90±2.25 hrs. The conception rate of 57.14% (4/7) was recorded in the buffaloes treated with insulin+Ovsynch (G-2), 50.00% (3/6) in the buffaloes treated with insulin alone (G-1) and 40.00% (2/5) in the group of buffaloes treated with Ovsynch alone (G-3). No significant difference was observed amongst all the treatment groups in terms of rate of induction of oestrus and time interval required for the induced oestrus.
