Description Caseinophosphopeptides (CPPs) are bio-active peptides (BAPs), play a vital role in immunomodulation. Studies were conducted to isolate, characterize BAPs derived
from buffalo milk proteins, their incorporation into weaning foods to explore hagocytic activity and lymphocyte proliferation in enhancing immunomodulatory property. Storage stability of weaning food was also studied.

Neutrase enzyme at an E:S ratio of 1:25 was used to achieve 10 percent Degree of Hydrolysis to produce BAPs of _-Casein and _-lactoglobulin. The higher degree of hydrolysis was observed for neutrase enzyme than trypsin. BAPs from neutrase hydrolysates of these protein fractions showed higher yield and molecular weight than BAPs from trypsin hydrolysate. HPLC analysis of BAPs revealed higher peak retention time for hydrophilic peptides.

Animal experimentation concluded that, the BAPs of _-casein are more promising in enhancing the immunomodulation in mice as it showed higher macrophage percentage (76 percent), phagocytic index (4.39), total leucocyte and lymphocyte count when compared to BAPs of _-Lactoglobulin with macrophage (58 percent) and phagocytic index (4.01). Storage stability of weaning food containing BAPs of these protein fractions revealed almost similar free fatty acid content and same degree of proteolysis compared to control. Hence, added BAPs did not affect the storage stability of weaning foods. This indicated that the BAPs can be safely incorporated into weaning foods without any adverse effects with good shelf life.
