The present study was undertaken with the objectives of seroprevalence, isolation,
molecular characterization of CSFV isolates of Karnataka and expression of TLR7 and
TLR8 in CSFV infected pigs. A total of 517 serum, 113 blood and 31 tissue samples were
collected from suspected field cases of CSF in different districts of Karnataka. The mean
prevalence of CSFV antibodies in serum samples for the 20 districts was 33 per cent
(173/517) with southern Karnataka having the higher prevalence. A total of 40 blood and
17 tissue samples were positive by RT-PCR. Among 113 blood samples, only 20 samples
were CSFV positive by antigen ELISA. A total of 15 CSFV isolates were isolated on cell
culture with two isolates as CPE producing isolates. Phylogenetic analysis based on E2,
NS5B and 5‟UTR sequences showed that all the 26 CSFV isolates belonged to subgroup
2.2. Comparison of phylogenetic analysis of CSFV isolates based on E2, NS5B and
5‟UTR sequences showed that, the most reliable classification was obtained with
sequence data from the NS5B and full length E2 (1119 nt) region. Analysis of deduced
amino acid sequences of full length E2 of 22 isolates revealed that, there were no changes
in conformational structure, virulence pattern and epitopes of E2 protein. There was
increased expression of TLR7 and TLR8 in 12 spleen tissues varied from 2 to 30 fold and
in 11 blood samples varied from one to 51 fold. The decreased expression of TLR7 and
TLR8 in 4 blood samples varied from 4 to 6 fold.
