The present study was planned for detection of PPRV from clinical samples using real time PCR assay. In present study 13 clinical samples (8 blood, 3 nasal swab, 2 tissue samples) as a representative from suspected sheep and goat were collected from Nagpur and Mouda tahsil in Nagpur district of Maharashtra. Detection of PPRV isolates was carried out using conventional method like RT-PCR as well as Real time PCR assay of M gene using SYBR green method. Conventional RT-PCR assay revealed that among these 13 clinical samples, 6 samples (3 blood samples, 1 nasal swab and 2 tissue samples) were found positive for PPRV indicating 46.15 % positivity. However, Real time PCR assay of M gene using SYBR green method revealed that all samples were positive for PPR indicating 100 % positivity. From the result it has been revealed that the Real time PCR of M gene using SYBR green assay has the advantages over the conventional RT-PCR and found to be rapid, sensitive and a suitable screening tool in addition to the earlier available techniques for confirmatory detection of PPR viruses in the clinical specimens of PPR suspect cases. |
