Description Maintenance of genetic purity and identification of the variety is utmost
importance in Seed production programmes. In order to maintain genetic purity,
Characterization is essential to carryout scientific seed production and
certification to enforce proper quality control and to promote the seed trade in the
present scenario. The current methodology utilized in seed production and
certification is Grow-out test which is time consuming since it involves detailed
observation of morphological characters. Moreover, most of the morphological
characteristics of varieties are common and influenced by environment. Hence
morphological characters alone can not be used for varietal identification. At this
juncture varieties can be identified by taking some of the morphological
characters and where ever the characters are common or of little difference we
should take the help of chemical tests or electrophoretic polymorphism.
In the present study, an attempt was made to characterize and identify
sixty four Greengram genotypes based on morphological characteristics
(Anthocyanin colour on hypocotyl, stem colour, stem pubescence, Seed colour
leaf size, pod colour, seed colour, seed coat luster, seed shape and size), reaction
of seed to different chemicals (NaOH, KOH) and seedling response to GA3
treatment and electrophoretic banding pattern of total soluble seed protein. Each
and every genotype could be distinguished based on specific morphological traits.
In order to determine the varietal identification, genetic purity assessment
and varietal characterization flowcharts have been developed based on
morphological characteristics.
Seed and seedling growth response to added chemicals viz., NaOH, KOH
and GA3 was also useful for grouping of genotypes.
Presence or absence of specific electrophoretic banding pattern of soluble
seed protein profile is useful for distinguishing each genotype from other .
Electrophoretic banding pattern of total soluble seed protein in each genotype was
unique and proved to be reliable and quick method for varietal identification.
