Description ABSTRACT
In the present investigation the genetic integrity of micropropagated plants of Jaspi-
Prunus rootstock was assessed using RAPD, SSR and ISSR markers. A reproducible protocol
has been developed for in vitro propagation. Nodal buds were used as an explants for in vitro
establishment of cultures of the rootstock. The sterilized explants cultured on MS medium
supplemented with 3mg/l GA3 and 0.75mg/l BA gave best results for in vitro shoot
multiplication. DNA from fresh, young and healthy leaves of 23 in vitro grown samples (15
samples from last four year micropropagated cultures and 8 samples from the last one year
micropropagated cultures) and a mother plant was isolated following CTAB method (Doyle
and Doyle, 1987) with some modifications. Out of total 35 primers used, 34 primers were
able to amplify the genomic DNA. A total of 11 RAPD primers were used and 10 primers
gave 38 monomorphic scorable bands with an average size range from 3000bp – 100bp.
Further, when SSR analysis was carried out, all 10 SSR primers gave amplification with 13
clear monomorphic bands of size range 3000bp – 800bp. Similarly all 14 ISSR primers gave
amplification with a total of 39 monomorphic bands of size range 5000bp-600bp. Thus on an
average 2.64 bands were amplified per primer. The three marker techniques were found to be
reliable to assess the genetic integrity of micropropagated Jaspi- Prunus rootstock.
