The present investigation was undertaken with an objective to assess seedling vigour traits in rice genotypes and identifying SSR markers for seedling vigour related traits. The material for investigation comprised of 47 different varieties, two high and two low vigour genotype. The experiment was laid out in a Completely Randomized Design with three replications at the Department of Seed Science and Technology, College of Agriculture, Acharya N.G. Ranga Agricultural University and molecular analysis was carried out at Directorate of Rice Research, ICAR, Rajendranagar, Hyderabad. Significant differences were observed for all the seedling vigour traits viz., seedling length, shoot length, root length, seedling dry weight, germination and vigour index. Highest germination percentage was recorded in Vandana, Savitri, Prasanna, Shalivahana, Vikas and Tulasi. High shoot length was recorded in Sabita, Vandana, Dinesh, Lalat, Swarandhan, Vasumati and Shalivahana. Similarly, highest seedling length was observed in Dinesh, Purnendu, Sabita, Vandana, Vivekadhan 62 and Shalivahana. Among all the genotypes, Dinesh, Purnendu, Sabita, Vandana, Vivekadhan 62 and Shalivahana showed highest root length and dry weight was showed higher in CSR 23, IR 64, DRR Dhan 39, PR 113, NDR 8002, NDR 359 and Sabita. Based on vigour index trait, eight genotypes were high in vigour index viz., Swaradhan, Tulasi, Vivekadhan 62, Dinesh, Sabita, Purnendu, Shalivahana and Vandana. The correlation studies revealed positive and significant association among all seedling vigour related traits. Vigour index was positively correlated with shoot and root length, seedling length, germination and seedling dry weight. The normal distribution for shoot length and seedling dry weight, seedling length and root length and skewed distribution for vigour index and germination percentage was observed. High heritability coupled with high GAM was observed for seedling vigour, root length and seedling length. Eight QTLs viz., qGR-1, qGP-6, qGI-11 RZ448, RZ395 qFV-3-2, qFV-5-2 and qFV-10 related to seedling vigour related traits were selected in the study. A total of 30 SSR markers were analyzed with a total of 51 genotypes of which 11 markers showed polymorphism. Among these 11 polymorphic SSR markers, highest phenotypic and genotypic correlation for seedling vigour traits was exhibited by RM11114 and RM20608. From the eight QTLs selected, six QTLs viz., qGR-1, qGP-6, qFV-3-2, qFV5-2 and qFV-10 were found to be present in different genotypes. The genotypes showed multiple QTLs viz., Dinesh has four QTLs viz., qGR-1, qFV-3-2, qGP-6 and qFV-10 and Pooja has two QTLs qFV-3-2 and qFV-10. Similarly, Sabita has two QTLs viz., qGR-1 and qFV-10 and Vivekadhan 62 has two QTLs qGR-1 and qFV-10. The present study concluded that the genotypes which showed high vigour were due to the presence of these putative QTLs associated with different seedling vigour traits and the markers identified may be useful in Marker Assisted Selection (MAS) for seedling vigour related traits and to develop molecular profiles pertaining to seed quality attributes. |
