Description |
A total of 1,000 drinking water samples from rural water supply, submersible pumps and hand pumps were analyzed bacteriologically by BIS 10500:1991 and BWTK. Epidemiological surveillance showed the maximum potability of tap drinking water samples (83%), followed by hand pumps (62%) and submersible pump (35%). Physiochemical studies showed acidic pH 6.3-7.0 in 26.4%, while 83.2% of drinking water samples showed TDS within the permissible limit. In drinking water samples, occurrence of Aeromonas hydrophila 76%, Yersinia enterocolitica 61 %, Listeria spp., 65%, Campylobacter spp., 63% and Clostridium spp. in 58% were observed. Heterotrophic plate count of submersible pump (76.8%), hand pump (47.8%) and taps (42.7%) were above the permissible limit (>500). The statistical analysis showed occurrence of emerging pathogens is independent of faecal coliforms, E.coli. An isolate characterized morphologically and biochemically was sequenced using 16s rRNA submitted in NCBI USA (Accession no GU596499). Scanning Electronic Microscopy of A. hydrophila depicted blunt ended, rod, size 1.17-2.24µM X 0.60µM. All the isolates showed MAR indices >0.2. Virulence marker test Crystal violet, Congo-red, Lipase, Autoagglutination and Deoxyribonuclease (DNase) were positive for presence of virulence plasmid. Maximum production of capsular polysaccharide was observed at 37°C, A3 31.64+0.12µg and 35.76+0.09 µg at 28°C and 37°C respectively, while minimum production by A1 strain, 9.62+0.03 µg and 16.05+0.02 µg at 28°C and 37°C respectively. Maximum expression of lipopolysaccharide production has been observed at 28°C by A1 strain 2.87+0.06µg and 2.53+0.03 µg at 28°C and 37°C respectively and minimum lipopolysaccharide production observed by MTCC strain i.e. 1.17+0.15 µg and 1.37+0.11 µg at 28°C and 37°C respectively. A3 strain showed the highest hydrophobicity. Highest MIC of sodium hypochlorite (4%) required is 1 ppm with contact time of 1 hour.
