Description The ability to distinguish and identify parental lines and its hybrid
is fundamental for the operational aspects of seed certification and seed
trade. An experiment was conducted to distinguish rice hybrid (KRH-4)
form its parental lines, using seed morphological characters, rapid
chemical tests, biochemical and molecular markers during 2012-13.
Among the rapid chemical tests, modified phenol test with copper
sulphate was found effective in differentiating the MSN36 (R) from
CRMS32A (A) and KRH4 (H). Further KOH and NaOH test was useful in
identification of MSN36 but needs refinement of protocol. However, not
much significant response has been noticed due to added chemicals like
GA3, Kinetin, 2, 4-D. The zymogram of total soluble seed proteins
revealed that regions E (30 to 18 kD), F (18 to 14 kD) and G (less than
14kD) were useful in distinguishing the parental lines and hybrid based
on relative intensity of bands. Among the tested isozymes, alcohol
dehydrogenase banding pattern found little useful in differentiating
parental lines and hybrid based on the intensity of bands. Out of 24 SSR
primers, 15 primers revealed polymorphism between parental lines and
hybrid. RM202 and RM204 are the polymorphic primers which can be
multiplexed for hybrid purity assessment. Among the polymorphic
markers of KRH-4, most of them were common to KRH-2, DRRH-1,
DRRH-2, CORH-1, CORH-2, CORH-3 hybrids. However, RM 1385 and
RM21 are found to be very unique markers for KRH-4. SSR primer
RM202 can be effectively used for hybrid purity assessment in KRH-4
seed lots also.
