Description ABSTRACT
Bacopa monnieri (L.) Pennell, commonly known as “Brahmi” is an important
medicinal herb of the family Scrophulariaceae. It has main importance as tonic for nervous
disorders and mental diseases. It helps in improving intelligence, memory and learning
ability. In the present work, a protocol for micropropagation of selected genotype of Bacopa
monnieri (L.) Pennell has been standardized. Axillary buds and leaves were used as explants
for in vitro plant regeneration studies in Bacopa monnieri (L.) Pennell. The explants were
sterilized with 0.2 per cent (w/v) bavistin and 0.1 per cent HgCl2 and maximum per cent
(80.95%) of uncontaminated cultures was achieved when 15 min treatment of 0.1 per cent
(w/v) bavistin was given followed by 2 min treatment of 0.1 per cent (w/v) HgCl2. Axillary
shoot buds showed best in vitro establishment (87.50%) on MS medium supplemented with
0.5 mg/l BAP and 0.5 mg/l Kn. Best in vitro shoot multiplication was obtained on MS
medium supplemented with 1.0 mg/l BAP and 0.5 mg/l Kn. Callus induction from leaf
explants was also carried out and it was found that MS medium supplemented with 1.0 mg/l
2, 4-D and 0.5 mg/l BAP was best for callus formation (53.33%). Leaf explants showed both
direct and indirect regeneration and MS medium supplemented with 6.0 μ mol/l BAP was
found to be best for shoot regeneration. For in vitro rooting, MS medium supplemented with
0.4 mg/l IAA was found to be the best medium (93.33%). Regenerated plantlets were
successfully acclimatized by using cocopeat followed by a mixture of sand: soil: FYM in
ratio 1:1:1.
