Description Field populations of Diamondback moth(DBM) infesting cabbage and
cauliflower in and around Hyderabad was used in the F2 screen for detecting the
presence of resistance alleles confirming resistance to Cry1Ac, cartap
hydrochloride and novaluron. From the 303 founder females only 27 produced
isofemales progeny of F2 larvae for screening the resistance alleles against
Cry1Ac, cartap hydrochloride and novaluron using leaf dip bioassay at their
respective discriminative dosages. Eleven F2 larvae belonging to five isofemale
lines survived in the F2 generation after bioassay and were re-evaluated for
confirming the presence of resistance alleles and were proved as false positives.
None of the F2 survivors confirmed the presence of resistance alleles in their
single pairs suggesting the absence of resistance alleles or confirming the
presence of SS genotypes.
The estimated frequency of resistance alleles in the present study using
Bayesian inference was 8.6x10-3 and associated variance was 6.93 x 10-5. The
95% Credibility Intervals for this isofemales was 2.1x10-4 to 3.3x10-2 (these are
the lower and upper limit for the expected frequency of resistance alleles) with
an experiment-wise detection probability of 52 per cent. No resistance allele
confirming resistance to the toxin and chemicals tested was detected in the
present study. A further sampling of few more isofemales lines would suggest
the implication of refugia concept for successful implementation of Bt cabbage
and Bt cauliflower cultivation in coming years.
