ABSTRACT :
A Competitive enzyme linked immunosorbent assay (C-ELISA) has been standardized for seromonitoring the vaccine response to haemorrhagic septicaemia (HS). Hyper immune serum against whole cell antigen of P.multocida P52 strain was used in the standardization of C-ELISA and in the characterization of immunogenic outer membrane proteins by Western blot immunoanalysis. The OMPs were extracted by a sarkosyl method and characterized by using SDS-PAGE. Five major polypeptide bands of MW 90 to 25 kDa were observed. Immunoblotting revealed that the polypeptides of MW 47,36 and 30 kDa were the major immunogens.
C-ELISA was standardized using both capsular and OMP antigens to measure the immune response in vaccinated animals. The efficacy of C-ELISA to differentiate between vaccinated and unvaccinated animals was evaluated by using sera from 70 animals vaccinated with HS alum precipitated vaccine. All the 70 post vaccinated sera were positive for HS antibody by both the
antigen, where as 3 samples out of 70 prevaccinated sera were also found positive. The mean ELISA titres in the post vaccinated sera were significantly higher than prevaccinated sera (P
