Description The anticoagulant, bromadiolone is widely used to control rodent populations effectively. Development of resistance to bromadiolone has been reported in several countries, but there is no such report from India. The present study was aimed to first determine the toxic doses of 0.005% bromadiolone bait against house rat, Rattus rattus, standardize blood clotting response (BCR) test and then to distinguish susceptible and resistant individuals of R. rattus based on the standardized methodologies and molecular characterization of VKORC1 gene. A total of 100 rats of both sexes collected from seven different poultry farms in district Ludhiana, Punjab were screened for development of resistance. Rats were fed on doses equivalent to LD 50 (2.10g/100g bw and 3.67g/100g bw for male and female rats, respectively) and twice the LD50 of 0.005% bromadiolone bait. Before dosing and after 48h of dosing, blood of rats was collected to record prothrombin time (PT), international normalized ratio (INR) and percent coagulation activity (PCA). Rats having PT less than 65 s, INR less than 4.0 and PCA more than 20 were suspected to be resistant. Overall, 36 rats out of the total collected were suspected to be resistant. Nucleotide sequencing of VKORC1 gene of one susceptible and two suspected to be resistant rats revealed no single nucleotide polymorphism. None of the sample under study showed complete nucleotide homology with previously reported nucleotide sequences of R. rattus. Therefore sequences of VKORC1 gene of R. rattus under study can be considered to be novel sequences that have not been reported earlier.
