The present investigation was undertaken with an objective to assess seedling vigour traits in rice genotypes, identify SSR markers for seedling root related traits and study association of genomic SSRs with seedling root traits and vigour. The material for investigation comprised of 47 rice varieties, comprising high, medium and low vigour genotype. The experiment was laid out in a Completely Randomized Design with three replications at the Department of Seed Science and Technology, College of Agriculture, Acharya N. G. Ranga Agricultural University and molecular analysis was carried out at Directorate of Rice Research, ICAR, Rajendranagar, Hyderabad. Significant differences were observed for all the seedling vigour traits viz., hundred seed weight, electrical conductivity, germination, shoot length, root length, seedling length, root shoot ratio, seedling fresh weight, seedling dry weight and vigour indices. Three genotypes BPT 5204, Kalinga III and Sampada had low seed weight, while the rest of the genotypes have medium to high seed weight. Of all the genotypes, Bharani Deep, IR 28, Pant Dhan16, Shabagyadhan, Suraksha, Vandana, VL Dhan82 and VL Dhan 208 showed low EC values. Highest germination percentage was recorded in Akshadhan, RP 2421, IR 28, Sahbhagyadhan, Sasyasree, Swarna, Vandana, VL Dhan 208 and Moroberekan. High mean germination time was observed in Heera, Krishnahamsa, RP2421 and Vandana. Bharani Deep, Dinesh, Narendra 118, Sahbhagidhan, Sasyasree and VL Dhan 221. High shoot length was observed in Dinesh, Heera, Jaldidhan, Narendra118, PNR 381, Sabita, Vandana, and VL Dhan 221. Similarly, highest root shoot ratio was found in Adithya, BPT 5204, Govind, NDR 97, Pant Dhan 16, Sahbhagidhan, Sasyasree, Sampada and Swarna. Highest seedling length was observed in Bharani Deep, Dinesh, Sabita, Sahhbagidhan, Narendra118, and VL Dhan 221. Among all the genotypes, Akshadhan, Dinesh, Jaldidhan, Moroberekan, Nagina 22, Suraksha and VL Dhan 208 recorded highest fresh shoot weight and Adithya, NDR 97, Sasyasree, Swarna and Vikas recorded high fresh root weight. Fresh seedling weight was showed higher in Akshadhan, Dinesh, Jaldidhan, Moroberekan, Nagina 22, Suraksha and VL Dhan 208. Dinesh, Jaldidhan, Moroberekan, Nagina 22, Suraksha and VL Dhan 208 recorded high seedling dry weight. Highest seed vigour index I was found in Bharani Deep, Dinesh, Sabita, Sahbhagidhan, Sasyasree, VL Dhan 208 and VL Dhan 221, where as high SVI II was noted in Dinesh, Jaldidhan, Moroberekan, Nagina 22, Nipponbare, Suraksha, VL Dhan 208 and VL Dhan 221. Of all physiological seedling traits root shoot ratio, root fresh weight, seedling dry weight and seed vigour indices showed high GCV per cent while root and shoot length, root shoot ratio, root fresh weight, seedling dry weight and seedling vigour indices recorded high PCV per cent. Among all the characters, highest heritability was recorded for seedling fresh weight. Based on vigour index trait, eight genotypes were high in seed vigour indices viz., Dinesh, Jaldidhan, Moroberekan, Nagina 22, Nipponbare, Suraksha, VL Dhan 208 and VL Dhan 221. The correlation studies revealed positive and significant association among all seedling vigour related traits, except for electrical conductivity and root shoot ratio. SVI II showed high positively correlated with germination, root and shoot length, seedling length, fresh root weight, fresh shoot weight, seedling fresh weight, seedling dry weight and SVI I. The normal distribution for root length, seedling length, root fresh weight and shoot fresh weight and skewed distribution for electrical conductivity, final count, mean germination, SVI I and SVI II was observed. High heritability coupled with high GAM was observed for SVI II. A total of 30 SSR markers analyzed for 15 QTLs in 47 genotypes, 16 markers showed polymorphism. Among these markers, highest phenotypic and genotypic correlation for seedling vigour traits was exhibited by RM351 and RM44. From the 15 QTLs selected 11 QTLs were found to be present in different genotypes. Some genotypes showed multiple QTLs viz., Moroberekan has nine QTLs, Nipponbare has six QTLs, VL Dhan 208 has five QTLs, DRR 38 has four QTLs. Three QTLs were present in Pantdhan16, Tulasi and Vandana. Two QTLs were present in Sampada, Anjali, Vivekadhan 62, Swarna, Dinesh. Final evaluation of high vigourous genotypes identified with phenotypic data and root QTLs by controlled deterioration test showed that Suraksha was the best genotype with high seed longevity period. The present study provided an overview of the phenotypic varience of the rice genotypes for seed vigour traits and their genetic diversity for root traits. There was large range of similar values for some root traits in related cultivars as microsatellites provide greater confidence for the assessment of polymorphism. The information about genetic diversity of these cultivars will be very useful for proper selection of parents and rice breeding programs especially for gene mapping and eventually for the application of marker assisted selection (MAS). |
