Description The present investigation was carried out to isolate Bacillus thuringiensis
from soils of different regions of Karnataka state, molecular
characterization to identify lepidopteran specific B. thuringiensis isolates
and bioassay of B. thuringiensis isolates against Diamondback Moth (P.
xylostella). A total of 20 soil samples were collected from different regions
of Karnataka state. Based on colony morphology 100 B. thuringiensis like
isolates were isolated from 16 soil samples. After microscopic studies
(gram staining, endospore staining and crystal staining) and biochemical
tests (catalase test, starch hydrolysis and casein hydrolysis) only 39
isolates were characterized as B. thuringiensis. Total DNA of these
isolates was extracted by following the procedure given by Hardwood and
Cutting, 1990 with modifications, PCR was carried out by using
lepidopteran cry gene specific primers. Out of 39 isolates only 20 isolates
showed amplification with lepidopteran cry gene specific primers, Among
these 14 isolates amplified with cry1 gene specific primer, 5 isolates
amplified with cry2 gene specific primer and 1 isolate amplified with both
cry1 and cry 2 gene specific primer. All 20 isolates which harboured
lepidopteran specific cry genes were used for bioassay against 2nd instar
larvae of Diamondback Moth. Isolate Bt-4 recorded maximum mortality
of 93.33 per cent which was lower than the standard strain HD1
(96.67%). This isolate was found to have both cry1A and cry2 genes.
Eight isolates showed more than 80 per cent mortality.
