Description ABSTRACT
The present investigation entitled “Studies on variations in seed progeny of Stevia rebaudiana
Bertoni” were conducted during 2010-2014 in the laboratory and experimental farm of Department of
Forest Products, UHF, Nauni-Solan (H.P.) with the objectives of (i) to study the extent of active
content variation in seed raised population and their correlation with morphological markers ii) To
identify superior genotypes. The studies were conducted under four experiments namely, (i) Studies on
inheritance of morphological characters and sweetener compounds in crosses between two known
genotypes (A×B) (ii) Studies on variation in morphological and sweetener characters in an open
pollinated seed raised population (iii) Evaluation of distinct strains (iv) RAPD and ISSR analysis for
molecular variation in selected morphotypes. Qualitative and quantitative morphological features were
observed on lamina shape, leaf tip, leaf base, leaf margin, plant height, number of main shoots/plant,
collar diameter etc. Chemical studies were focused on the percentage content of stevioside and
rebaudioside-A. Observations on fresh leaf yield/plant was recorded to assess the yield potential in
selected plant types. Genetic diversity among the selected plant types was assessed through RAPD and
ISSR markers. F1 and F2 progeny raised through crosses between already known genotypes A and B
revealed considerable variation with regard to morphological as well as chemical parameters. Often
plants with similar morphological features revealed different stevioside as well as rebaudioside-A
content thereby indicating no correlation between any morphological and chemical parameter. The
stevioside content ranged from 0.64 per cent to 16.18 per cent and rebaudioside-A content ranged from
0.00 per cent to 8.55 per cent. Chemotypes with higher rebaudioside-A content (F219, F220, F221, F222
and F223 ) and low stevioside content were identified (plant numbers F26, F27 and F218). Most of the
higher rebaudioside-A and low stevioside content plants were observed in F2 progeny of A and B.
Hybrids developed between selected parent lines did not reveal any better plant types in terms of
biomass and/or chemical content. A non significant average fresh leaf yield ranged from 531.50g to
697.50g/plant. RAPD and ISSR analysis revealed that plant numbers OP-3 and OP-15 were formed to
be independent genotypes with the former being least disease susceptible and later with higher
rebaudioside-A than stevioside content. Similarly for other plant types analysed, the similarity index
was 0.50 or less than 0.50 indicating the selected plant types being genetically diverse. Overall the
significant achievement of the study is identification of plant numbers F219, F220, F221, F222, F223 and
OP-15 with higher rebaudioside-A than stevioside content which is a most desirable character for
commercialization of this species.
