• FMDV recombinant protein and BSA were used for rasing antibodies in rohu. FMDV recombinant protein was more immunogenic than BSA Both FMDV linked and BSA linked column were found to be efficient to purify rohu Ig in active form Both purified proteins from FMDV sera and BSA sera showed antibody activity in a mouse antiserum based Immunodot In a non reducing SDS-PAGE, only BSA Ig gave a protein b and with molecular weight 850 KD similar to that of other fish IgM In SDS-PAGE analysis FMDV Ig and BSA Ig showed heavy and light chain protein bands of 59 and 46 KD and 85, and 23 KD respectively Both FMDV Ig and BSA Ig were used to develop anti rohu Ig MAbs In western blot, MAbs raised against both FMDV Ig (F4g12, F4H9 and F2D9) and BSA Ig (B4G12 and B2D1) were found to react with 85 KD protein of BSA Ig . Except the MAb F4H9 allthe MAbs reacted to 59 KD protein of FMDV Ig Results of non reducing SDS- PAGE, SDS-PAGE and Western blot reveals that BSA Ig is a tetrameric Ig and FMDV Ig is either a low molecular weight Ig or a degraded form of rohu Ig. MAbs F4G12 and F2D9 were found to be most sensitive in an Immunodot assay Reactivity of the MAb F4G12 with both sera and mucus of rohu in an Immunodot indicates that serum and mucus Ig shares common epitopes Cross reactivity of MAbs F4G12, F2D9 and B4g12 with Cyprinids and other fishes indicates that rohu Ig shares epitopes with other fish Igs. MAb F2D9 could be successfully employed to detect antigen specific immunoglobulins in rohu in a post immunization and post infection scenario |
