The present investigation entitled “GENETIC DIVERSITY AND PROTEIN PROFILING STUDY IN PUMPKIN (Cucurbita moschata Duch. ex. Poir.)” was carried out at Vegetable Research Centre, G. B. Pant University of Agriculture and Technology, Pantnagar, District-Udham Singh Nagar (Uttarakhand) during summer 2006 and summer 2007. The experiment was laid out in randomized block design with 24 genotypes collected from different part of country. The objectives of present study was to estimate the extent of variability, correlation studies, path analysis, genetic diversity and protein profiling pattern through SDS-PAGE of the different genotypes. The observations were recorded on 22 characters viz., vine length, no. of primary branches per vine, node of 1st male flower appear, node of 1st female flower appear, days to anthesis of 1st male flower, days to anthesis of 1st female flower, sex ratio, polar diameter of fruit, equatorial diameter of fruit, polar diameter of seed cavity, equatorial diameter of seed cavity, flesh thickness, fruit weight, fruits per plant, fruit yield per plant, 100 seed weight, seeds per fruit, total fruit yield per hectare, fruit shape, fruit surface, rind colour and flesh colour. Analysis of variance revealed highly significant differences for all the characters. Based on the findings of present investigation PPU-146, PPU-199, PPU-46, PPU-195, PPU-260 and PPU-197 were few top yielders. High GCV and PCV was observed for days to anthesis of 1st male flower, days to anthesis of 1st female flower, fruit yield per plant, total fruit yield per hectare, node of 1st male flower appear. The high heritability coupled with high genetic advance was obtained for total fruit yield per hectare, fruit weight, vine length, node of 1st female flower appear, node of 1st male flower appear, days to anthesis of 1st male flower, 100 seed weight, equatorial diameter of seed cavity, polar diameter of seed cavity, polar diameter of fruit and equatorial diameter of fruit. Total fruit yield per hectare had highest positive and significant phenotypic correlation with fruit weight, fruit yield per plant, equatorial diameter of fruit, flesh thickness, equatorial diameter of seed cavity and polar diameter of fruit. On the basis of Mahalanobis’ D2 analysis, the 24 genotypes of pumpkin were grouped into 5 clusters in year 2006 and 4 cluster in year 2007. The cluster I had the highest genotypes (11 in year 2006 and 15 in year 2007), followed by II (7 in year 2006 and 7 in year 2007), III (4 in year 2006) while there were two single variety cluster (cluster IV and V) in year 2006 and (cluster III and IV) in year 2007. Seed protein profiling of 24 genotypes could resolved into a total of 17 bands distributed in 6 district zones. Genotypes CM-71, CM-83, PP-113, PPU-138 and PPU-144 were included under one cluster which was most distinct from PPU-39 with 35.29% similarity showing greater divergence. |
