Description The present study was conducted on 120 healthy male mice aging 7-8 weeks to investigate the effects of single (one day) and multiple exposures (6 and 24 days) to poultry barn air of floor and cage-housed system. The animals were divided into four groups viz. three treatments and one control for each experiment. Among the treatments groups, Group 1 was exposed for single day, Group 2 for 6 days (Monday to Saturday) and Group 3 for 24 days (4 weeks, Monday to Saturday). At the end of exposure period, half animals from each group were challenged with LPS @ 100µl/ animal and the remaining with NSS @ 100µl/ animal. Single and multiple exposures of 6 days to cage-housed system resulted increased expression of mRNA of TLR 4 and IL-1β mRNA compared to floor-housed system. The expression of TLR4 was down regulated after multiple exposures of 24 days. There was increased expression of IL-1β mRNA after multiple exposures of 24 days. There was almost 10 and 50 folds increase in the expression of TLR9 mRNA following single exposure and exposure followed by LPS challenge to cage-housed system respectively. Multiple exposures of 6 days down regulated the expression of TLR9 mRNA but 24 days exposures resulted increase in the expression. Single exposure to cage-housed system showed signs of acute inflammation and multiple exposures of 6 days resulted maximum histopathological alterations.
