Description Yellow mosaic disease caused by a Begomovirus is an important production
constraint in lima bean cultivation in Karnataka. Survey conducted during 2013-14 to
assess the yellow mosaic disease incidence in lima bean growing areas of selected
districts of southern dry zone of Karnataka revealed the occurrence of the disease in the
range of 25.3 to 81.1 per cent. Among the districts surveyed, highest average disease
incidence was recorded in Chikkaballapura (72.03%) and least incidence was recorded in
Ramanagara (31.95%) district. Lima bean yellow mosaic virus (LYMV) was
successfully transmitted to French bean, horsegram, pole bean, ring bean and soybean
through whiteflies (Bemisia tabaci). The virus was detected by PCR using viral coat
protein (CP) and movement protein (MP) specific primers, which amplified ~1000bp and
~900bp product corresponding to the CP and MP gene of the virus. The sequence of the
CP and MP shared maximum nucleotide identity of 98% and 99% with Horsegram
yellow mosaic virus (HgYMV) isolates infecting French bean (FB) and horsegram from
Bangalore. The DNA-A component (2.7kb) of the virus was also amplified and
sequenced. The sequences showed maximum nucleotide identity of 99% with HgYMVBangalore:
FB isolate. Phylogenetic analysis of the sequences of LYMV genomic
components viz., DNA-A, MP and CP genes revealed that LYMV-GKVK isolate
clustered with HgYMV infecting horsegram and French bean isolates of Tamil Nadu,
Srilanka and Bangalore, which indicates that the virus causing lima bean yellow mosaic
disease is a strain of HgYMV.
