Description In present study on “Biochemical composition of allantoic and amniotic fluid in
different stages of pregnancy in goat” total 50 gravid goat uteri were collected from
Nagpur municipal slaughter house immediately after slaughter of goats. After exposing
the amniotic and allantoic sacs, allantoic and amniotic fluid was collected and
immediately transferred to – 200c refrigerator for further biochemical analysis. According
to crown rump length of fetus in the gravid uterus the samples were divided from Group
– I to Group – V.
In allantoic fluid of gravid goat uterus, glucose and creatinine concentration was
13.44±2.29, 30.75±6.65, 25.11±4.73, 18.40±1.69,15.12±1.79 and 0.36 ± 0.18, 3.23 ±
0.61, 6.81 ± 1.05, 11.16 ± 2.01, 14.13 ± 1.64 mg/dl, respectively in Group – I, II, III, IV and V. In Group – II, III, IV and V allantoic fluid, uric acid concentration was 1.05±0.15,
3.2±1.29, 1.15±0.09 and 1±0.13 mg/dl, respectively. Urea nitrogen concentration of
allantoic fluid from goat gravid uterus varied from 2.30±2.48, 23.74±3.05, 18.54±2.29,
19.16±2.86 and 11.91±0.98 mg/dl, respectively in Group – I, II, III, IV and V. Total
protein concentration of 0.36±0.04,1.33±0.33,1.13±0.26,1.03±0.18 and 1.04±0.06 mg/dl,
respectively was observed in Group – I, II, III, IV and V allantoic fluid in present study.
Allantoic fluid in Group – II, III, IV and V gravid uteri goat shows chloride concentration of
59.29±5.57, 25.06±3.63,16.73±2.64 and 13.28±2.52 mmol/l, respectively. Sodium
concentration of 106.25±16.31, 94.7±29.67, 77.3±12.74, 131.05±39.03 and
82.55±8.01mmol/l, respectively was recorded in the allantoic fluid from gravid goat uterus
in Group – I, II, III, IV and V. Average potassium concentration of 2.27±1.22, 3.53±2.17,
0.6±0.47, 7.66±3.94 and 0.72±0.32 mmol/l was observed in allantoic fluid of gravid goat
uteri from Group I, II, III, IV and V, respectively. Glucose, creatinine, uric acid, urea
nitrogen, total protein, chloride, sodium and potassium concentration shows significant
difference between different age groups for their concentration in the allantoic fluid from
gravid goat uteri.
Glucose, uric acid and urea nitrogen concentration of 5.96±1.23, 29.49±6.91,
36.79±4.07, 21.63±4.14, 23.07±5.17; 0, 1.9±0.44, 1.06±0.04,1.08±0.16, 0.7±0.18 and
1.59±1.77,10.67±1.75,10.06±1.37,11.88±1.26 and 6.21±0.79 mg/dl, respectively was
observed in the amniotic fluid from gravid goat uteri in Group – I to V. Average
concentration of creatinine in amniotic fluid from gravid goat uteri ranged from 0.04±0.03
to 4.88±0.49 mg/dl between groups.
In Group – I, II, III, IV and V amniotic fluid from gravid goat uteri, average total
protein concentration of 0.33±0.02, 0.25±0.07, 0.23±0.03, 0.36±0.13 and 0.26±0.04
mg/dl were observed, respectively. Average chloride concentration remains constant and
showed non significant difference for amniotic fluid of gravid goat uteri in Group – III,
Group – IV and Group – V. Amniotic fluid showed average concentration of
125.15±9.70, 139.25±28.45, 124±18.20, 147.8±31.55,102.8±8.32 and 3.29±0.89,
41.94±35.61, 4.7±0.97, 7.38±2.46 and 1.531±0.50 mmol/l sodium and potassium ion,
respectively in Group – I to V gravid goat uteri, respectively.
In gravid goat uteri from Group – I to Group – V, glucose, creatinine concentration
was significantly higher in allantoic than amniotic fluid whereas uric acid concentration
was significantly increased in amniotic than in allantoic fluid. Urea nitrogen concentration
significantly increased in Group – I in amniotic than in allantoic fluid, its concentration
gets significantly increased in allantoic fluid than in amniotic fluid with the advancement of pregnancy. Total protein concentration was significantly increased in allantoic fluid of
pregnant goats from Group – I to Group – V. Chloride ion concentration significantly
increased in amniotic fluid. Significantly higher sodium ion concentration was present in
amniotic fluid than in allantoic fluid from Group – I to V. Amniotic fluid potassium ion
concentration increases significantly in Group – II to IV gravid goat uteri, whereas in
Group – V, it increased significantly in allantoic fluid.
Allantoic and amniotic fluid volume of 76±12.22, 122±29.37, 161±27.32,
202±38.61, 193±8.17 and 59±5.15, 120±17.27, 292±38.71, 291.5±35.02 and 310.5±9.44
ml, respectively was recorded in the gravid goat uteri from Group – I to V. pH of amniotic
fluid is comparatively higher than that in allantoic fluid. Average crown rump length of
fetus from gravid goat uteri was 7.08±0.67, 15.29±0.82, 27.53±1.15, 35.01±0.64 and
43.73±0.95cm for Group – I to V, respectively.
