tomato (Lycopersicon esculentum Mill.) genotypes using RAPD

Description Abstract
The present studies on ‘‘Genetic characterization of tomato (Lycopersicon
esculentum Mill.) genotypes using RAPD” was carried out to study the genetic
relationship among various genotypes. Besides the proposed objective, work on Simple
Sequence Repeats (SSR) was also done. The aim of this study was to investigate genetic
relationship among different genotypes of tomato. 21 tomato genotypes namely 1-(EC-
28373), 2-(Pepsi92), 3-(SolanVajr), 4-(EC-521086), 5-(EC-528374), 6-(101), 7-(UHF-
517), 8-(UHF-562), 9-(UHF-659), 10-(UHF-521), 11-(EC-528367), 12-(AC-402), 13-
(110264), 14-(AL-14), 15-(UHF-562), 16-(UHF-506), 17-(UHF-571), 18-(UHF-505), 19-
(UHF-55), 20-(UHF-571), 21-(EC-521074) were selected for the study. A total of 25
primers were tried to generate RAPD profile, out of these reproducible patterns were
obtained with 15 primers. A total of 72 bands were obtained out of which all were
polymorphic and 3 were unique bands and in case of SSR studies 9 primers were used
and total number of bands obtained were 33 with 3 unique bands. The percentage of total
polymorphism was 100% in both RAPD and SSR. Similarity index was computed based
on Jacquard coefficient and used for cluster analysis based on UPGMA. RAPD and SSR
technology could be useful for identification of different accessions as well as accessing
the genetic similarity among different accessions of tomato.
