Description ABSTRACT
Random amplified polymorphic DNA (RAPDs) and simple sequence
repeats (SSR) were used to evaluate the clonal stability of four year old
micropropagated shoot clusters of Merton I.793. Shoots multiplied under in vitro
conditions on suitable medium were categorized on the basis of three
morphotypes with minor differences. For assessment of genetic fidelity, DNA
was isolated from leaves of mother plant grown in the field and in vitro raised
shoot cultures of MI.793. Samples with good quality DNA were selected for
amplification reaction. Out of the 31 RAPD primers screened, 27 produced a total
of 130 scorable and distinct bands with an average of 3.65 bands per primer. 26
primers resulted in monomorphism across all the shoot clusters while, little
(2.3%) polymorphisim was obtained with a single primer where 3 bands were
absent in four shoot clusters. Of the 6 SSR primers, 5 generated 9 bands with an
average of 1.8 bands per primer indicating complete monomorphism. The
banding patterns for each primer were highly uniform and comparable to the
mother clone from which the cultures had been established. DNA markers
revealed genetic similarities within micropropagated shoots and mother plant of
Merton I. 793. Our results show that micropropagation protocol for this rootstock
is reliable and emphasizes its utility for large scale propagation over a long period
of time, without much risk of genetic instability.
