Description Barley (Hordeum vulgare L.) is one of the world’s most important crop. Leaf blight
caused by Helminthosporium sativum Pam., King and Bakke has been known to occur on
Barley in Karnataka in an epiphytotic form. To know the possible factors for disease
resistance, various parameters like biochemicals, isozymes and DNA finger printing of
resistant genotypes, DWR 28 and PL 760 and susceptible genotypes, RD 2653 and RD 2508
were undertaken.
Resistant genotypes recorded higher total phenol, total protein, reducing sugar, total
sugar, chl ‘a’, chl ‘b’ and total chlorophyll, isozyme activity (peroxidase, polyphenol oxidase
and phenylalanine ammonia lyase) compared to susceptible genotypes which are possible
factors for disease resistance.
DNA finger printing clearly indicated the difference in resistant and susceptible
genotypes. The maximum polymorphism for both resistant and susceptible genotypes was
designated by primer OPA-05 and OPA-02 respectively.
Overall studies revealed that, higher biochemical content (total phenol, total proteins,
reducing sugars, total sugars, chl ‘a’, chl ‘b’, total chlorophyll and high isozyme activity
(peroxidase and polyphenol oxidase) and enzymatic activity (phenylalnine ammonia lyase)
are possible factors for disease resistance to barley leaf blight disease caused by H. sativum.
