Description Zinc deficiency is the second most spread nutrient deficiency worldwide. Rice is
the major source of all nutrients for the population in India but it is deficient in Zn. An
effort was made to study the effect of nucleotide variations in genes associated with Zn.
20 genotypes contrasting for grain Zn were selected and grown in pots at UAS, GKVK in
CRD, during Summer-2013. PCV and GCV were highest for number of tillers, number of
productive tillers, grain yield, root length, root number, root volume, grain Zn, shoot Zn
and root Zn. Highly positively significant correlation was observed for grain yield with
number of productive tillers and productivity per day per plant. Literature survey
revealed genes belonging to families like OsZIP, OsYSL, OsNRAMP are involved in
transporting Zn. Primers were designed specific to these genes. Single-marker analysis of
the primer profile at gel level revealed that OsZIPc was significantly associated to shoot
Zn. Sequencing the amplicons of primers OsNRAMP, OsYSL2b, OsZIP3b, OsZIP4a,
OsZIP5b and OsZIP6c revealed that variations were present in all these amplicons but,
nucleotide changes at seven positions of OsZIP6c were significantly associated with
shoot Zn content and nucleotide changes at four positions of OsZIP5b were significantly
associated with grain Zn. Although OsZIP5b was monomorphic at gel level, nucleotide
variations were significantly associated with grain Zn content. Hence any monomorphic
marker may have information as much as a polymorphic marker. The markers identified
in this study can be used for improving the grain Zn in rice.
