In the present study, various aspects of biology of Puntius sarana sarana were studied over a period of one year. A maximum weight and (pooled) recorded was 68.45 and 78.0 g for males and females, respectively, whereas the maximum length (pooled) recorded was 21.5 and 21.8 for males and females, respectively. The sex ratio varied between 1:1 and 1: 4 (M: F), with an average sex ratio of 1:2.16. In females the GSI varied between 0.55 and 7.4, while the male GSI ranged between 0.23 and 2.8. The absolute fecundity varied from 1,932 to 11,511 eggs per female and the relative fecundity ranged between 49,294 and 1,55,204. Based on the colour, shape, size and macroscopic examination of gonads, six maturity stages (Stage - Stage VI) were observed in female and five maturity stages (Stage - Stage V) were recognised in male. The histological examination of ovaries revealed the presence oogonia, chromatin nucleolar oocyte and perinucleolar oocyte, yolk vesicle stage, of vitellogenic oocytes, ripe oocyte, etc. in the ovaries and spermatogonia, spermatocytes, spermatids and spermatozoa etc. in the testis. The egg and embryonic development proceeded normally showing distinct blastodisc formation, 2, 4, 8, 16, 32, 64, 128, 256 cell stages, followed by morula, blastula, early gastrula, middle gastrula, late gastrula, yolk plug, pea shape, bean shape, embryo indication and advanced. The average length of the newly hatched larva was 1.90 ± 0.002 mm. The early larval development also proceeded normally. The yolk sac was completely absorbed in 58-72 hours after hatching. The mean genetic distances between different species within the same genus Puntius was 0.22. A percentage genetic distance of 22.6% between P. lithopidos and P. sarana subnasutus, 13.0% between P. lithopidos and P. lithopidos and 6.5% between P. sarana sarana and P. sarana subnasutus was evaluated. The pairwise divergence rang was evaluated and it ranged between 8 and 16% in the COI partial sequences among the five species. This suggests that P. carnaticus, P. pulchellus, P. sarana sarana, P. lithopidos and P. sarana subnasutus can reasonably be regarded as five different species. |
