Thermal springs are a major source of potentially significant but untapped microbial diversity encoding novel genes. Metagenomics bypasses the need for isolation or cultivation of microorganisms as it deals with direct isolation of nucleic acids from environmental samples. The present study was designed to analyse the profile of microbial community inhabiting hot water spring Tattapani in Himachal Pradesh, situated at an altitude of 656m above sea level. The spring water has temperature 63°C and pH 7.5. It was rich in sulphur. Metagenomic DNA was isolated by using RapidWater ™ DNA Isolation kit. MGUF-MGUR, U789F-U1053R and FBI-RPII 16S rRNA gene specific primers were used to amplify partial 16S rRNA gene. In-silico analysis showed presence of both Archaea and Bacteria in the spring water. Bacteria included Clostridials bacterium, Thermus scotoductus, Thermodesulfobacterium, Pseudomonas aeruginosa, Hydrogenophilus thermoluteolus, alpha and beta proteobacterium, uncultured bacterium, marine bacteria plankton etc. Archaea phylum Euryarcheota was also identified using culture independent approach. Attempts were also made to culture and characterise microbes by culture dependent approach. These were identified as Anoxybacillus flavithermus, Westillopsis sp. and Nostoc punctiforme.
