Description |
Peste des Petits Ruminants (PPR) is an acute and highly contagious disease of small ruminants characterized by high fever, necrotic stomatitis, catarral inflammation of ocular and nasal mucosa, pneumonia and severe enteritis with high morbidity and mortality. The current understanding of peste des petits ruminants virus (PPRV) pathogenesis has been mostly assumed from the closely related morbilliviruses as the available information on this aspect is meager. The lack of a suitable laboratory animal model has impeded progress of understanding how PPRV causes disease and the role of cell mediated immunity against this virus. In the present study, rabbit was used as a laboratory animal model for studies of immunokinetics of PPRV. PPRV Sungri/96 strain was used to infect rabbits via I/N and I/M routes. All the rabbits remained apparently healthy following infection and no clinical signs including thermal reaction were observed. Regardless of the route of inoculation, PPRV RNA was detected intermittently in nasal, rectal and blood samples of infected rabbits mostly between 4 and 8 dpi employing real time qRT-PCR. However, attempts to isolate PPRV from the afforsaid samples in Vero cells were not successful. The cell mediated immune response of rabbits against PPRV and Phytohemagglutinin (PHA) was assessed in vitro by lymphocyte proliferation assay (LPA) using MTS dye. The mean proliferative response against PHA was evident and significantly higher (P |
