Description Safflower (Carthamus tinctorious L.) is a valuable oil seed crop, which can contribute
for meeting the edible oil demands of the country. Concerted efforts are required to improve
safflower for high oil yield potential in order to make it attractive for farmers and the
industry. Enhancement of oil yield requires simultaneous improvements in seed yield and oil
content. Molecular mapping would enable identification of genes or genomic regions
contributing for oil content and their exploitation in molecular breeding programmes, which
has not been attempted in safflower to date. In this context, the present study was conducted
to detect quantitative trait loci (QTLs) associated with seed oil content using SSR markers in
safflower. A set of 94 F2:3 progenies produced from a cross between A-1 (low oil content,
~26%) and EC-755673-1 (high oil content, ~38%) was used as a mapping population. The
population was evaluated for oil content in a field trial at Rajendranagar farm of IIOR,
Hyderabad. Both the parents had at least 10% difference in oil content (in absolute terms) and
the F2:3 progenies exhibited quantitative variation (range 20.7%-39.4%). Polymorphism of the
parents, A-1 and EC-755673-1 was analysed at a total of 1107 SSR loci (in 3% agarose gel),
which resulted in identification of 54 polymorphic loci (5%), which included 13 already
mapped (anchor) loci and 41 unmapped loci.
The mapping population was genotyped with 54 polymorphic SSR primer-pairs in
agarose gel (3%) and polyacrylamide gel (6%) electrophoresis. The SSR alleles were scored
in Mendelian fashion as 1 for A-1 allele, 2 for heterozygous and 3 for EC-755673-1 allele.
Chi-square test was carried out to check if the segregation of SSR markers followed
Mendelian inheritance pattern. Out of 54 SSR makers, chi-square value of 36 markers were
not significant suggesting that these markers followed Mendelian segregation of 1:2:1 in the
F2 population and the remaining 18 markers showed deviation from the expected segregation.
