In the present study, physico-chemical parameters were analyzed in soils from Rann of Kutch experiencing high levels of salinity. Analysis showed wide range of variation in pH, Electrical Conductivity, potassium and the Na+ concentration, Na+/K+ ratio, available nitrogen and organic carbon content. The principal component analysis revealed two principal components and the longest distance from the point of origin was for Na+, K+, Na+/K+ ratio and EC which suggested their major role in relation to the presence of cyanobacteria. Regression analysis indicated a significant and positive correlation between EC and Na+ content. However, a significant and negative correlation was found between EC and number of cyanobacteria. Similar negative correlation was observed for the number of cyanobacteria and available nitrogen.The cyanobacterial isolates were examined microscopically for morphological attributes Cyanobacterial forms identified morphologically belonged to Anabaena, Nostoc (order Nostocales), Phormidium (order Oscillatoriales) and Westiellopsis (Stigonematels). The extracted and quantified DNA from cyanobacterial isolates was subjected to 16S rDNA gene amplification and the amplified product was used for sequence analysis and the BLAST analysis established the homology in terms of % similarity
