Description |
The present study was carried out on the infundibulum, ampulla and isthmus part of oviduct of sixty goats. The samples collected from abattoir immediately after slaughter of goat were categorized into two groups of follicular and luteal phase and processed for histomorphological, histochemical and ultrastructural studies. The various staining methods were followed for histological and histochemical studies. The microscopic and micrometrical observations were carried out after processing the samples for paraffin sections. For ultrastructural characterization, transmission and scanning electron microscopic studies were carried out.The histological studies revealed primary and secondary mucosal folds in all parts of oviduct during both phases of estrous cycle. Tertiary branching of mucosal folds was found only in infundibulum and ampulla. In infundibulum and ampulla, height of epithelium was more in follicular phase than in luteal phase, whereas in isthmus, it was more in luteal phase. In some secretory cells of infundibulum and ampulla, prominent cytoplasmic protrusions were found protruded in the lumen. The height of ciliated as well as secretory cells showed increasing trend from infundibulum to isthmus in both phases of estrous cycle. Lamina propria was found merged with submucosa as no muscularis mucosae was present in all parts of oviduct. The muscle layer was more thick and prominent in isthmus than in infundibulum and ampulla. At the apical border of epithelium of infundibulum and isthmus, PAS reaction was intense during luteal phase .The apical epithelial border showed intense and weak AB-PAS reaction in infundibulum and ampulla in both phases of estrous cycle for presence of neutral and acid mucopolysaccharides respectively.The present transmission electron microscopic study revealed secretory and ciliated cells as two distinct cells within the epithelium of all parts of oviduct. In infundibulum and ampulla, secretory cells showed prominent apical cytoplasmic protrusion into the lumen during both phases of estrous cycle. In the secretory cells of ampulla, numerous secretory granules of different size and density were noticed. Release of secretory granules from the secretory cells was noticed in all parts of oviduct in both phases of estrous cycle. In the apical cytoplasm of ciliated cells, fibrous granules, basal bodies and electron lucent vesicles were noticed in all parts of oviduct during both phases of estrous cycle. In the ciliated cells of all parts of oviduct, rootlets were extending in the apical cytoplasm from basal bodies. In the scanning electron microscopic study, mucosa was thrown into longitudinal primary folds in all parts of oviduct. In the isthmus, numerous shallow crypts were observed in the basal area of pockets. The infundibulum and ampulla were extensively ciliated with many bulbous processes of secretory cells located among the ciliated cells during follicular phase. During both phases of estrous cycle, sticking of secretory material over the luminal surface was noticed in infundibulum and ampulla part of oviduct. |
