Description In the present investigation, in vitro studies were carried out with a prime
objective of developing a suitable protocol for high frequency regeneration from
various explants viz., hypocotyls, cotyledonary leaf and primary leaves of five
selected sesame genotypes viz., JCS-402, Hima, RT-125, Rajeshwari and Swetha.
Studies were also conducted to determine the transformation frequency in terms of
transient GUS expression percentage in direct explants (hypocotyls, cotyledonary
leaves, primary leaves) and callus derived from them.
Between the hormonal combinations (BA + NAA and TDZ + NAA),
MS medium supplemented with BA + NAA was found better for callus induction
from hypocotyls, cotyledonary leaves and primary leaves than MS medium
supplemented with TDZ + NAA. Among the BA + NAA treatments, 2 mg/l BA +
0.2 mg/l NAA was found better for callus induction.
MS medium supplemented with 4 mg/l BA and 0.2 mg/l NAA and MS
medium supplemented with 4 mg/l SA and 0.5 mg/l NAA were found better for
inducing maximum caulogenesis and rhizogenesis. Among the five sesame cultivars,
JCS-402 followed by Swetha, Rajeshwari, Hima and RT-125 was found better for
callus induction, while JCS-402 followed by Hima, RT-125, Rajeshwari and Swetha
was found better for caulogenesis. RT-125 followed by JCS-402, Rajeshwari, Hima
and Swetha was found better for rhizogenesis.
Transformation frequency in terms of transient GUS expression percentage
was determined in direct explants and their respective calli of five sesame genotypes
using particle inflow gun. Primary leaves were found better as target tissue for
particle bombardment among the direct explants as they gave highest transient GUS
expression percentage than callus derived from them. The studies suggests
suitability of direct explants for transformation studies of sesame using particle
inflow gun.
