The present study was undertaken to note the in vitro germinability and direct shoot
organogenesis on cotyledonary node segments in six genotypes. The factors standardized
were pre-treatment soak of dry seed in 50 mg/l GA3 for 24 h which gave the best
germination on full-strength B5 medium supplemented with 2.0 mg/l BAP + 0.2 mg/l
NAA. Comparative in vitro seed germination in six genotypes, namely, JS 335, Pusa 9712,
Pusa 24, Pusa 9814, Line 3 and Dwight showed considerable variation. The earliest seed
germination was noted in JS 335 (3.6 days) followed by Pusa 24 (3.7 days). Germination
was highest in JS 335 (92.3%) followed by Pusa 9814 (83.3%), while the lowest
germination percentage was recorded in Dwight (10.5%). The cotyledonary node segments
were then excised and transferred for shoot proliferation. The in vitro shoot multiplication
was highest (4.53 shoots/ explant) in treatment comprising half-strength MS (macro) + fullstrength
MS (micro + organics) + 2.0 mg/l BAP + 2.0 mg/l kinetin followed by treatment
half-strength MS (macro) + full-strength (micro + organics) + 1.0 mg/l BAP + 1.0 mg/l
kinetin (3.57 shoots). These treatments also gave good shoot length, internode length and
minimum days for sub-culture (19.2 days). Addition of 1.0 mg/l GA3 to MS medium gave
sufficient shoot elongation (3.17 cm). The micro-shoots were then excised individually and
transferred to root medium comprising full-strength MS medium. The earliest rooting (7.43
days) was recorded in the treatment IBA (1.0 mg/l) + NAA (1.0 mg/l) compared to other
treatments, which also gave the highest percentage of rooting (94.07). The rooted plantlets
gave the earliest hardening, i.e. the glass jar with polypropylene (PP) cap (28.57 days)
compared to plastic pot with polythene cover (34.07 days). The glass jar with PP cap was
