Description An effective plant regeneration and gene transfer system was developed for Punica granatum L. cv. Kandhari Kabuli. an economically important species of the tropical and subtropical regions of the world due to its delicious edible fruits. pharmaceutical and ornamental usage. Calli and direct adventitious shoot buds were initiated from juvenile explants cxcised from 15 to 40 days old in invitro germinated seedlings. The best media for callus induction from cotyledon, hypocotyl internode and leafexplants were MS medium supplemented with 13.0 μM NAA and 13.5 μM BA. 13.0 μM NAA and 18.0 μM BA, 5.0 μM IBA and 90 μM BA, 8.0 μM NAA and 9.0 μM Kinetin. respectively. The highest percentage of callus was obtained from cotyledon (85.50) explants followed b) hypocotyl (79.67). internode (79.47) and leaf (75.48) explants. The calli thus obtained showed differentiation on MS medium supplemented with 9.0 μM BA and 2.5 μM NAA. Cotyledon derived callus showed the highest regeneration (81.97%) with 16.47 mean number of shoots followed by hypocotyl, internode and leaf derived calli. The regeneration and rooting potential of the callus showing high proliferation rate but no signs of differentiation decreased with increase in subculture passage. The direct adventitious shoot bud induction from cotyledon as well as hypocotyl explants lIas observed on MS medium supplemcnted with 9.0 μM BA. 9.0 μM Kinetin and 2.5 μM NAA when the cultures were maintained in continuous darkness for twO weeks and then transferred to 16-hour photoperiod. The maximum adventitious shoot bud induction was observed in cotyledon explants (71.17%) showing 6.17 shoots per explant followed by hypocotyl (60.63%) explants showing 5.67 shoots per explant. The adventitious shoots obtained were rooted on 1/2 strength MS ll1edium containing 500 mg l-1 activated charcoal. Eighty percent of plant lets were successfully established in earthen pots containing soil and sand (1:1).
