Huntington’s disease (HD) is characterized by selective death of the medium spiny GABAergic neurons in the striatum. Worldwide a large population of people suffers from this progressive degenerating disease and a lot more are at risk. Although the genetics of HD was known by the end of twentieth century the exact molecular mechanism leading to the selective striatal pathology is still a mystery. Till date no cure or preventive measures are available for the disease and the therapies available for the treatment arem mostly symptomatic. So the focus of HD research demanded a shift from molecular genetics to neurobiology with the hope that understanding the basic mechanisms of pathology will lead to newer therapeutic strategies. Mainly because of these reasons researchers are constantly involved in developing animal models that manifest the cardinal features of the disease that are reproducible and at same time quantifiable. The latest evidence of this effort was reflected in development of a transgenic monkey model of the disease. In the present study an animal model has been established by administering a mitochondrial neurotoxin 3-NP in rats and characterized to ensure its reliability and reproducibility in terms of behavioral, histopathological and neurochemical attributes. The model is then employed to study the basic molecular mechanisms that lead to the demise of the striatal cells. The status of proteasome, a principal muticatalytic protein degradation complex was also investigated under the pathological condition induced by 3-NP in this model. The outcome of the entire work is documented in this thesis. |
